Bone homeostasis is regulated and maintained by a tunable balance between bone-forming cells (osteoblasts) and bone-resorbing cells (osteoclasts). Under healthy physiological conditions, bone has several functions, including (a) protection of the central nervous system, (b) release of Ca2+ or other ions, (c) mechanical support for soft tissues, and (d) housing and support for hemopoiesis. At the macro and microscopic levels, the structure and mass of bone are regulated by the genetic blueprint, physical factors, and (bio)chemical factors which regulate bone homeostasis. The main aim of the studies presented in this thesis was to investigate the effect of physical and (bio)chemical niche conditions on (pre)osteoblast mechanoresponse and behavior. We first introduced the concept of niches in bone and muscle. Via mechanical interaction with their niche, osteocytes and MuSCs are subjected to mechanical loads causing deformations of membrane, cytoskeleton, and/or nucleus, which elicit biochemical responses and secretion of signaling molecules into the niche. The latter may modulate metabolism, morphology, and mechanosensitivity of the secreting cells, or signal to neighboring cells and cells at a distance. Thereafter, we found that mechanical loading changed the osteoblast cell surface area, cell body, and nucleus volume.
